'Master' proto-oncogene regulates stress-induced ovarian cancer metastasis

Jan. 29, 2013 ? Scientists at The University of Texas MD Anderson Cancer Center have discovered the signaling pathway whereby a master regulator of cancer cell proteins -- known as Src -- leads to ovarian cancer progression when exposed to stress hormones. The researchers report in the current issue of Nature Communications that beta blocker drugs mitigate this effect and reduce cancer deaths by an average of 17 percent.

Src (pronounced "sarc," short for sarcoma) is a proto-oncogene -- a normal gene that can become an oncogene due to increased expression -- involved in the regulation of cell growth and division. Known to be instrumental to changes that cause normal cells to go awry, this study sheds light on its functional role as a key molecular switch affecting a downstream signaling pathway that spurs disease progression.

Led by Anil K. Sood, M.D., professor in MD Anderson's Departments of Gynecologic Oncology and Cancer Biology, researchers found that noradrenaline (NA; a stress hormone) directly affects tumor growth and spread through beta-adrenergic (ADRB) receptors expressed on tumor cells. The study demonstrated that ADRB signaling leads to Src activation via a unique protein kinase A (PKA)-mediated mechanism, which is critical to the regulation of cellular activity and cancer metastasis. This is the first time that scientists have been able to show that ADRB receptors play a direct role in Src activation by this mechanism.

"When Src is triggered by stress, it works like a dam letting out water that causes a flood downstream. Src, like the dam, is a master regulator switch that causes a chain reaction in the cells," said Sood.

Based on existing findings of Sood's ongoing work exploring potential interventions against the effects of stress, the researchers examined data on outcomes of cancer patients treated with beta blocker drugs from the U.S. Food and Drug Administration Adverse Event Reporting System. They found that mortality in patients treated with a beta blocker was reduced by an average of 17 percent across all major cancer types. Moreover, they observed a nearly 15 percent decrease in mortality among patients with ovarian and cervical cancer.

Beta blockers, also called beta-adrenergic blocking agents, treat a variety of conditions, such as heart disease, high blood pressure, glaucoma and migraines. They act on the ADRB receptors, which are also found on the heart -- causing the heart to beat harder and faster under stress -- and are involved in maintaining blood flow.

When the ADRB receptors on cancer cells are activated, they set into motion a chain of events that leads to formation of new blood vessels that feed tumor growth -- a process known as angiogenesis. New blood vessel formation allows tumors to grow and spread more rapidly. Beta blocking agents stop this process.

"Prior to our work, the concept of stress hormones driving cancer growth was very new and only very limited information about the effect of beta blockers on cancer outcomes in humans has been available," said Guillermo Armaiz-Pena, Ph.D., instructor of Gynecologic Oncology and Reproductive Medicine and first author of the study. "This study provides incentive to further explore beta blockers as a possible supplement to traditional cancer therapies."

A Mystery Pathway Revealed

While NA -- the most abundant stress hormone in the ovary -- has been proven to modulate multiple cellular functions important for cancer progression, how it does so had remained a puzzle. Sood's team used a multi-step process to determine how the tumor microenvironment is disrupted by stress hormones.

First, the researchers exposed ovarian cancer cells to NA and identified a number of proteins altered by stress hormones. Using bioinformatics analysis, they narrowed potential mediators to Src.

A series of subsequent experiments designed to verify the biological roles of Src in promoting ovarian cancer tumor growth in response to stress hormones revealed the signaling pathway involved in NA-mediated Src activation. Specifically, they showed:

? PKA (also known as cAMP-dependent protein kinase) is the switch that "turns on" NA-induced Src activation; ? The signaling pathway occurs at a particular site on the cell known as S17; and ? This specific mechanism is key to mediating ADRB/cAMP/PKA-induced Src activation.

Building on the Stress-Cancer Connection For the past 13 years, Sood's research efforts have focused on the effects of chronic stress on cancer metastasis. The latest study helps form a more comprehensive picture on the impact of and biological mechanics of chronic stress on ovarian cancer, as well as the role of beta blockers in slowing disease progression. Previous studies have shown:

? Chronic stress triggers a chain of molecular events that protects breakaway ovarian cells from destruction, as heightened levels of the fight-or-flight hormones epinephrine and norepinephrine permit more malignant cells to safely leave the primary tumor -- a necessary step in metastasis and cancer progression.

? When mice with ovarian cancer are stressed, their tumors grow and spread more quickly, but the effect can be blocked using propranolol, a beta blocker commonly prescribed for heart disease.

Future research will focus on other biological mechanisms that may be affected by stress. Eventually, Sood hopes his studies will help identify the cancer patients most likely to benefit from beta blockers and other stress interventions. He is also looking at the impact of stress on other diseases, such as gastrointestinal disorders.

"This is a major step forward in understanding the biology and impact of stress on cancer progression and it opens the door to study drugs that could inhibit this unique signaling pathway," Sood said.

Reuters reports -- French-backed Malian troops searched house-to-house in Gao and Timbuktu on Tuesday, uncovering arms and explosives abandoned by Islamist fighters, and France said it would look to hand over longer-term security operations to African troops.

French and Malian troops retook the two Saharan towns in northern Mali virtually unopposed at the weekend after an 18-day French-led offensive that has pushed back the al Qaeda-allied militants into hideouts in the deserts and mountains.

Malian government soldiers were combing through the Niger River towns and their neighborhoods of dusty alleys and mud-brick homes. In Gao, they arrested at least five suspected Islamist rebels and sympathizers, turned over by local people, and uncovered caches of weapons and counterfeit money.

Residents reported some looting of shops in Timbuktu owned by Arabs and Tuaregs suspected of having helped the Islamists who had occupied the world-famous seat of Islamic learning, a UNESCO World Heritage site, since last year.

Fleeing Islamist fighters torched a Timbuktu library holding priceless ancient manuscripts, damaging many.

Chevron Paid $10 Million in Claims Following Refinery Fire

Chevron U.S.A. has paid approximately $10 million in claims to compensate area hospitals, affected community members and local government agencies since its August fire at a refinery in Richmond, Calif., according to an update by the company released this week.

* On Aug. 6, 2012, a fire broke out due to a piping rupture at the oil refinery in Richmond, the report stated. Due to a high level of sulfur dioxide released with the emission, residents of Richmond, San Pablo and North Richmond were advised to stay indoors until the fire was controlled.

* Other materials released into the air from the fire include methane, non-methane hydrocarbon, hydrogen sulfide and nitric oxides.

* The Contra Costa Health Services website reported that a large number of people sought medical attention at local emergency rooms, mostly for minor complaints of nose, throat or eye irritation and respiratory issues. Three individuals were admitted to the hospital, Chevron stated.

* Some minor employee injuries were also reported with the incident, including burns, abdominal discomfort, respiratory irritation, blisters and bruises, the company stated.

* Air sampling in the surrounding area on Aug. 7-8 showed that the concentrations of hydrogen sulfide, sulfur dioxide and carbon monoxide were below detection limits, the company stated.

* According to the company, a claims process was established in order to compensate community members for medical and property expenses that occurred because of the fire. As of Jan. 21, the company says about 23,900 claims have been initiated and it has paid $10 million in compensation.

* Chevron stated this week that its investigation of the incident is ongoing and that it is working with multiple governmental agencies to discover what caused the leak in the piping system that led to the fire.

* The Associated Press reported in early January that federal investigators believe that firefighters may have accidentally damaged the pipe while trying to put out a small flash fire that had been caused by a leak in the line. The larger damage in the pipe would have released more gas, and could have led to the larger fire.

* According to Reuters, Chevron -- the second-largest U.S. oil company -- is planning to bring the Richmond refinery back to full production this quarter. Even though it posted decreased refining input in the third quarter of last year because of the fire, the company's worldwide output was the was the highest it has been since mid-2011 at the end of the year.

Djokovic sets sights on elusive French Open

(AP) ? In years past, Novak Djokovic marked his victories at the Australian Open with rowdy late-night celebrations and bleary-eyed photo shoots the next morning in downtown Melbourne.

This year's win made history but inspired a more sober reaction.

After beating Andy Murray to become the only man to win three consecutive Australian Open titles, the No. 1-ranked player didn't feel the need to celebrate immediately.

Instead, he booked an early Monday flight home to start preparing for his next challenge: the clay courts of Europe.

The Serb has the Davis Cup next weekend and a few months away is the French Open ? the one major that has eluded him. Djokovic now has six Grand Slam tournament trophies, four overall from the Australian Open and one each from Wimbledon and the U.S. Open in 2011. He came close last year at Roland Garros, but lost in the final to clay-court master Rafael Nadal.

"Of course, I want to go all the way in the French Open," Djokovic said at his post-match news conference just after midnight.

His goal for the year is a big one, he said, when asked if he would choose a Roland Garros title over his No. 1-ranking.

"I'll take everything," the 25-year-old Djokovic said. "I have no reason not to be confident in myself."

Djokovic never lacked self-confidence and his dominating performance at the Australian Open showed just why.

The elite group Djokovic heads includes No. 2 Roger Federer, No. 3 Murray and Nadal, whose creaky knees caused him to sit out this tournament and yield his No. 4 ranking to David Ferrer. Djokovic beat Ferrer in an 89-minute semifinal he said he played "perfectly."

Federer, a 17-time Grand Slam-winner, Nadal, Djokovic and Murray have combined to win 33 of the past 34 majors.

"I have a great feeling about myself on the court at this moment," Djokovic said after beating Ferrer.

His 6-7 (2), 7-6 (3), 6-3, 6-2 victory over Murray on Sunday night showed his mental toughness and supreme fitness in a match that contained riveting rallies between two of the best returners in the game.

The win deprived Murray of his chance to capitalize on his breakthrough year in 2012, when he won an Olympic gold and his first major title at the U.S. Open.

"I'm full of joy right now," Djokovic said. "It's going to give me a lot of confidence for the rest of the season, that's for sure."

The season resumes next weekend with Serbia's Davis Cup tie against Belgium, which was why Djokovic flew home early so he could celebrate with the people closest to him.

"In life you don't get many opportunities to win Grand Slams. As a tennis player, that's a pinnacle of the ambitions and of the success," he said. "So I (will) try to enjoy it for a few days with the people I love the most ? family, friends and team."

Djokovic apologized to reporters for skipping Monday's traditional post-victory news conference.

"The main reason is because I want to get to Europe as quick as possible so I can be ready for the Davis Cup tie," said Djokovic, who led Serbia to its first and only Davis Cup title in 2010. "I hope I find your understanding for that."

After he wrapped up his media obligations, he went online at about 4:30 a.m. to post a note of thanks to his fans.

"My dear friends," he wrote in a personal blog post that he also tweeted. "(I'm) laying in bed now and thinking 'Novak, you are 4 times AO champion,' That's quite something, right? I will have to repeat it in my mind for a while to sink in."

"This is just the start of the year!" Djokovic wrote, ahead of catching his flight. "Plenty of things ahead of us, starting from tomorrow morning. ... Stay tuned."

Wall Street climbs at open on earnings

NEW YORK (Reuters) - Stock futures climbed on Friday and the S&P 500 was poised for an eighth day of gains, its longest winning streak in eight years, buoyed by rosy earnings from Procter & Gamble amid a backdrop of sturdy corporate results.

The equity market was also boosted by agreement in Washington to extend the government's borrowing power through mid-May, encouraging signs of recovery in the global economy, solid corporate earnings, and seasonal inflows into stocks.

Those factors helped the S&P 500 rally for a seventh day on Thursday to a five-year peak. Still, the index struggled to climb convincingly above 1,500, a level it surpassed briefly Thursday for the first time since December 2007.

"We are seeing a very broad-based rally and the ingredients are still in place" for gains to continue, said Steve Goldman, principal at Goldman Management in Short Hills, New Jersey. "This is the lift-off phase and it's still significant."

Procter & Gamble , the world's top household products maker, said quarterly profit soared past expectations and raised its sales and earnings outlook for the fiscal year. Shares were up 2.1 pct at $71.88 in premarket trading.

S&P 500 futures rose 3.6 point and were above fair value, a formula that evaluates pricing by taking into account interest rates, dividends and time to expiration on the contract. Dow Jones industrial average futures rose 32 points and Nasdaq 100 futures rose 9.75 points.

If the S&P 500 rises for an eighth day, it will be its longest winning streak since late 2004, when it enjoyed a nine-day run.

Pointing to a rotation out of bonds, U.S. 30-year Treasury bonds traded more than a point lower in price on Friday, with yields touching session highs at 3.10 percent.

"You have had more confidence from fund managers to provide more allocations to equity markets," which looked more attractive than bonds or cash, said Rick Meckler, president of investment firm LibertyView Capital Management.

Recent company earnings have been encouraging. Thomson Reuters data through early Thursday showed that of the 133 S&P 500 companies that have reported earnings so far, 66.9 percent exceeded expectations, more than the 65 percent average over the past four quarters.

Microsoft Corp reported lower quarterly profit on Thursday as Office software sales slowed ahead of a new launch, offsetting a solid but unspectacular start for its Windows 8 operating system and sending the company's shares down 0.8 percent in premarket trading.

Apple stepped up audits of working conditions at major suppliers last year, discovering multiple cases of underage workers, discrimination and wage problems. The shares, which fell 12 percent Thursday after disappointing earnings, edged up 0.5 percent to $452.90.

German business morale improved for a third consecutive month in January to its highest in more than half a year, providing further evidence that growth in Europe's largest economy was gathering speed after contracting late last year.

Echoing a more positive tone in Europe, ECB President Mario Draghi said on Friday he expects the euro zone economy to recover later this year, and that financial market improvements had not yet trickled into the general economy.

Europe's emigrants face red tape in Latin America

(AP) ? Geologist David Rodriguez and actress Cristina Pascual, two of the nearly six million Spaniards left jobless in the European recession, fled to Latin America last year, figuring their futures would be brighter in the booming economies on this side of the Atlantic.

Instead, they found themselves stuck, facing so many bureaucratic hurdles that their only option was to work illegally, for much lower wages. Without a work visa, they couldn't get a formal job. Without a job offer, no visa. And without a job and a visa, they had no way of securing an all-important tax-identification number, freezing them out of Chile's booming formal economy. Trying to bend the rules can result in deportation for the worker, and fines for the company.

Rodriguez and Pascual are among the many migrants watching this weekend as leaders from the European Union, Latin America and the Caribbean seek ways of eliminating the red tape that has made it so difficult for foreigners to bring their skills across borders.

The vast majority of migrants between the continents used to travel to Europe, but the trends flipped after 2010, when economic indicators began to improve in Spain and Portugal's former colonies. Now Spanish and Portuguese workers are arriving by the thousands each year, entering on tourist visas and job-hunting in Brazil, Chile, Mexico and Argentina. Spanish migrants with skills needed in the mining industry are particularly sought after in Chile.

But the bureaucracy is getting in the way.

Rodriguez, 32, graduated from college in Spain in 2010, and spent the next 28 months in a frustrating search for work in his field while he lived with his parents. Then a friend, architect Maria Moreira, posted a Facebook message saying she was migrating to Chile. He jumped at the chance to come along, but soon found himself stuck without authorization to work. Despite high demand for exactly the skills he could offer, it took four months of rejections before he found a low-paid internship that eventually led to a company's sponsorship for his paperwork.

Spain's national statistics service said more than 40,000 Spaniards abandoned the country in search of work during the first six months of last year, up sharply from the 28,000 during the same period in 2011. The same agency said this week that unemployment has reached a record 26 percent in Spain.

Reducing roadblocks is urgent for the leaders at this summit. The IMF forecasts the Latin American and Caribbean economies to grow 3.6 percent this year even as Europe retreats 0.2 percent. The continents' economies are inexorably tied to each other, with EU countries representing 43 percent of Latin America's international trade. Making it easier for workers to move to where the jobs are can help all these countries, in part by increasing the remittances people send home to their families.

Chile isn't unique in the demands it places on foreign workers. Brazil and Argentina are famous for their red tape, and lacking EU citizenship, many Latin Americans haven't been welcomed into Europe's job market, either.

The summit agenda includes fostering "best practices" for lowering barriers to work. One example: representatives of 400 universities on both sides of the Atlantic met in Santiago this week to create a "common space for higher education," with the goal of standardizing the degrees and certifications awarded throughout both regions.

The agenda also advocates equal treatment for citizens of all nations, a sore point in the former colonies of Europe. While EU citizens can enter any country in the region on tourist visas, Latin Americans have been humiliated in Europe's airports, interrogated and sent home even though they said they complied with the entry rules.

"Welcome to a better world," is how Chile's President Sebastian Pinera greeted his Spanish counterpart Mariano Rajoy, a tongue-in-cheek phrase that resonates on all sorts of levels for travelers between both regions.

Rodriguez and Pascual can testify that their welcome was difficult at best during their first months in Chile.

Without formal work and a visa, they couldn't get the Chilean identity numbers known as RUTs, which are vital for all aspects of life. Without a RUT, you can't get a telephone line, buy a cellphone, obtain a credit card, open a bank account, or make hundreds of other transactions in Chile.

Pascual, a 38-year-old actress, spent five months in low-paid, under-the-table jobs before she got help from the Spanish embassy, where she now works as a cultural representative.

"The bureaucracy here is terrible," Pascual says. But now she sounds contented, and spends much of her time supporting other Spaniards who keep arriving in Chile. "Many people are coming, so we help them a little, because it's very bureaucratic here, very complicated, and they get desperate."

    Council mulls possibility of a tax hike to offset state cuts, avoid ...

    Sheffield Lake City Council?s Jan. 15 meetings were dominated by financial discussions, some aimed at increasing city revenues in the future and others aimed at devising ways to cope with current losses of revenue, in the form of some sort of tax increase. Possibilities included 1-mill property tax levies, an income tax increase or a decrease to the current 0.5-percent income tax credit provided to city residents who work outside the city.

    In the Finance and Audit Committee meeting before council?s work session, finance Director Tammy Smith spelled out the drastic need for some new revenue to offset a total of $280,000 in revenue loss from the state of Ohio (including cuts to local government and library assistance that helped to balance the state?s budget, and the phased-out business personal property tax), which will be aggravated by reduced real estate tax revenue in the wake of last year?s lowering of property values by the Lorain County Auditor?s office.

    Smith opined that the city?s smaller than normal budget appropriations could suffice ??if there were no need to pay any overtime wages to anyone and if the city could go an entire year without incurring the kind of expenses demanded to deal with such things as water main breaks, etc. In addition to that unlikely scenario, Smith said, was an immediate need to acquire three new police cruisers and at least one new ambulance, a new generator and other new equipment for the fire station.

    ?We could put 1-mill equipment levies for the safety departments on the ballot, but that could pose a hardship on our senior residents on fixed incomes,? Smith said. ?Or we could ask for a one-quarter of 1 percent income tax increase, or for a one-quarter of 1 percent reduction to our one-half of 1 percent income tax credit and reduce that to a quarter of 1 percent. We had 1-mill safety equipment levies in the past, but dropped them in 2003 when we asked the voters to raise the income tax a quarter-percent to help finance our street maintenance and improvement program. Either of those income tax changes could raise about $280,000. That would replace the state cuts ??and get us back to our 2009 level.?

    Future revenue-raising possibilities discussed included the creation and implementation of a Community Investment Corporation (CIC) to attract new business to the city. Law Director David Graves said a CIC would have powers not available to the city itself. Like a port authority, a CIC would have the power to borrow or lend money to jump-start a new business and could even enter into contracts without advertising for bids. Westlake?s Crocker Park development, Graves added as an example, was financed with a loan from the Toledo Port Authority.

    Graves said that council discussions of creating a CIC to attract investors to the Shoreway Shopping Center began in the summer of 2009, but it was never implemented. Graves said the structure of a CIC would include (in addition to the mayor and two other administration members) a five-member board of directors to be appointed from among city residents. The CIC was never implemented, mainly for the reason (as several members of council pointed out) that it could prove very difficult to find five qualified residents willing to volunteer their time and expertise.

    Planning Commission member Edward Rinderknecht asked council to consider creating an ordinance to address the appearance of new commercial buildings proposed on Zone B-1 or B-2 commercial lots that are surrounded on three sides by R-1 residential zones.

    ?I know it would be too late to have any effect on the Dollar General project at Lake and Harris, but we need to consider future development,? Rinderknecht said as he distributed copies of a Lakewood ordinance that influenced the appearance of the Marathon gas station on Lake Road in Lakewood. ?That station now blends in beautifully with the residential neighborhood,? Rinderknecht said.

    Council President Rick Rosso asked members to mull the possible tax issues and decide which way to go in time to put it into ordinance form before a Feb. 6 filing deadline, which must be met in order for the Lorain County Board of Elections to put a tax issue on the May primary ballot.
    ?If we?re going to do this, we?re going to have to put an ordinance together and approve it at our Jan. 23 regular meeting to meet the filing deadline,? Rosso said.

    Dimon apologizes to shareholders for "whale" loss

    (Reuters) - JPMorgan Chase & Co Chief Executive Jamie Dimon apologized to shareholders for the $6 billion loss caused by the so-called "whale" trade, calling it a "terrible mistake," but said the bank has moved on and is still highly profitable.

    "If you're a shareholder of mine, I apologize deeply," Dimon said at a presentation at the World Economic Forum in Davos, Switzerland. "But we had record results and life goes on."

    Despite a $6.2 billion loss from bad trades in JPMorgan's chief investment office last year, the bank still managed to earn a record $21.3 billion in 2012.

    JPMorgan Chase is the largest U.S. bank, with $2.36 trillion in assets as of December 31. Its chief investment office has since been restructured and traders and executives involved with the bad trade - referred to as the "whale" trade after the nickname of a London-based trader involved - were dismissed.

    After an internal review, Dimon's bonus was cut in half to $11 million for 2012.

    Back in 2004, Illinois attorney Sean Smoot's organization was the first labor and police group to endorse then-U.S. Senate candidate Barack Obama. To close the loop, he said, Smoot came back to Washington this weekend to celebrate the start of Obama's second term as President.

    "We're thrilled to have someone as president who's not only from our state, but someone who is a friend to law enforcement," said Smoot, the chief legal counsel for the Illinois Police Benevolent and Protective Association.

    Smoot and his wife, Teresa, got almost the exact same seats for the Inauguration ceremony as they did four years ago. But the weather was much milder this time, and so was the crowd control and organization for getting people in and out, he said.

    As for the energy? "The scene looks the same," Smoot said. The weekend for him and his wife included a dinner with Senator Dick Durbin (D-Ill.), the Illinois social ball on Saturday, and a rooftop party at the Willard InterContinental hotel to watch the parade.

    Smoot said that the police association has enjoyed strong relationship with Obama's administration. He also serves as treasurer of the National Association of Police Organizations, which works with the U.S. Department of Justice to help secure funding for policing programs.
