UFC 160: Cain Velasquez Ready for Junior dos Santos After Antonio Silva Victory

At UFC 160, Cain Velasquez proved that his first-round knockout of Antonio Silva a year ago wasn?t a fluke and that he?s more than ready for round No. 3 against Junior dos Santos.

Who Will Win Velasquez-dos Santos III?

Who Will Win Velasquez-dos Santos III?

  • Cain Velasquez

  • Junior dos Santos

Velasquez defended his UFC Heavyweight Championship by knocking out Silva in Round 1 once again on Saturday. UFC President Dana White announced in the post-fight press conference that the champ?s next opponent would be the only fighter to ever defeat him, dos Santos.

"No-brainer," said White of the Velasquez-dos Santos matchup according to Ben Fowlkes of USA Today. ?If there was ever a trilogy, that's it right there. If you look at the way both fights went?Junior destroys (Velasquez) in the first fight, Cain destroys (dos Santos) in the second fight?I can't wait to see the third fight.?

In November of 2011 in UFC on Fox, dos Santos shocked Velasquez, knocking him out with punches 1:04 into the first round. Not only was that Velasquez?s first loss, but it was only the second time he failed to knock out his opponent as a professional mixed martial artist. A year later, Velasquez got his revenge, reclaiming his heavyweight title belt from dos Santos via unanimous decision.

In Velasquez-dos Santos III, expect a result closer to their most recent bout.

Dos Santos is more than talented enough to reclaim the title belt. ESPN ranked him as the No. 2 heavyweight in the world, behind only Velasquez. Health should be a concern, though.

Following their first bout, dos Santos suffered from a potentially fatal condition, rhabdomyolysis. WebMD describes this condition as ?a serious syndrome due to a direct or indirect muscle injury? which ?results from a breakdown of muscle fibers and release of their contents into the bloodstream,? according to Kevin Iole of Yahoo! Sports. He also endured major injuries against Frank Mir and prior to his first fight against Velasquez.

Injuries didn?t hold dos Santos back as he knocked out Mark Hunt in the third round of their UFC 160 bout, but his health concerns are starting to add up so it will difficult to unseat the scorching Velasquez.

Jolie's aunt succumbs to breast cancer - Rappler.com

LOS ANGELES, USA - Angelina Jolie's aunt has died of breast cancer, it was reported Sunday, less than two weeks after the Oscar-winning actress announced that she had undergone a preventative double mastectomy.

The Hollywood entertainment website E! News cited Jolie's uncle, Ron Martin, as saying that his wife, Debbie -- the younger sister of Jolie's late mother -- died early on Sunday at Palomar Hospital in Escondido, California.

"Angelina has been in touch throughout the week and her brother Jamie has been with us, giving his support day by day," Martin was quoted as saying on E!'s website.

"They both loved Debbie very much and although Angie is not able to come right now she has sent her love and support which was very nice."

Jolie, 37, revealed in an article in the May 14 edition of The New York Times that she chose to undergo surgery to minimize the risk she might develop breast cancer due to the inheritance of a "faulty gene."

The actress's partner and fellow screen star Brad Pitt led worldwide praise, declaring Jolie heroic, followed by her doctors, other stars and thousands of supporters, who took to social media to praise her openness.

Jolie and Pitt have six children -- three adopted and three biological. The actress's mother, Marcheline Bertrand, died of ovarian cancer at the age of 56 and passed on the gene, BRCA1, that put Jolie at higher risk.

Jolie's doctors estimated she had an 87 percent risk of developing breast cancer and a 50 percent risk of ovarian cancer, prompting her to take action to cut the chances that she might die at a young age.

The actress said in the newspaper article that her likelihood of developing breast cancer is now just five percent, though she still runs a relatively high risk of contracting ovarian cancer.

Jolie, respected for her humanitarian work overseas with the UN, said she was speaking out to help other women understand their options, and also to urge governments in lower-income countries to provide the health care women need.

READ: Breast cancer and Angelina's choice

"I started with the breasts, as my risk of breast cancer is higher than my risk of ovarian cancer, and the surgery is more complex," she wrote. "I can tell my children they don't need to fear they will lose me to breast cancer."

Cancer campaigners cheered Jolie but warned women against rushing out to be tested for the gene mutation that threatened the actress's life.

Not only is the BRCA1 mutation rare in the female population, they said, but it is also expensive to test for -- Jolie paid $3,000 -- at a US laboratory that controversially claims patent rights on the gene. - Rappler.com

Hon Hai, Mozilla to launch mobile device running on Firefox

TAIPEI (Reuters) - Taiwan's Hon Hai Precision Industry Co Ltd will team up with Mozilla and launch a mobile device that runs on the U.S. company's Firefox operating system, a Mozilla spokeswoman said, as Hon Hai looks to diversify its client base away from Apple.

The Taipei-based Mozilla spokeswoman declined to provide details on the device that the companies plan to unveil on June 3, but Focus Taiwan, citing an "industry insider", reported that it would likely be a tablet.

Hon Hai and Mozilla declined to comment on the report.

Hon Hai, a contract electronics maker that draws an estimated 60 percent of its revenue from assembling gadgets and other work for Apple Inc, saw its net profit ease last quarter as sales of iPhones and iPads slowed.

Most of the mobile devices that Hon Hai manufactures run on Apple's iOS and Google's Android systems. Hon Hai said it would add Firefox to its product line for clients.

Mozilla, the non-profit organization that evolved from Netscape, said at Barcelona's Mobile World Congress in February that it was working with manufacturers Alcatel (TCL Communication Technology), LG Electronics and ZTE Corp to build the first Firefox OS devices, with Huawei Technologies to follow later in the year, all powered by the Qualcomm Snapdragon mobile processors.

Call of Duty: Ghosts DLC will launch first on Xbox One

Despite its multiplatform status, Call of Duty is almost synonymous with Xbox. It's no surprise then, that Microsoft's new platform, the Xbox One, will be the first next-gen system to digest Call of Duty: Ghosts' DLC. This isn't unprecedented, of course -- the Xbox 360 also boasted a first-out-the-door policy with the series' DLC. Still, a big score for Microsoft, one significant enough to serve as the closing announcement of its Xbox One launch event. Activision's publishing CEO Eric Hirshberg went on to reveal the game's first official trailer, assuring fans that Ghosts would fun at a steady 60fps. "It will be the best Call of Duty game we've ever made," he promised, showing slides comparing the new game's Xbox One visuals against previous games in the series. Hungry for more? Patience, friend. E3 is still a few weeks away.

Interview: Josh Ritter on fatherhood, touring, The Beast in Its Tracks ...

Credit: Dean Chalkley

Last month, I spoke to Josh Ritter over the phone during a short break from touring behind his new album, The Beast in its Tracks. That conversation informed a feature article on Josh and the new album, which you can read here. Now we?re posting an edited transcript of the interview for you; check it out below.

Before you do though, you can enter to win tickets to Ritter?s show at The Trocadero tomorrow night. More details on the giveaway are here.

?

The Swollen Fox: First off I?d like to say congratulations on becoming a father ? Beatrix Wendylove Ritter [is] almost five months old [now six]. How?s she doing?

Josh Ritter: Thank you. She?s awesome. She?s awesome, she?s amazing, she?s practicing laughing right now. So, pretty great.

TSF: Where did you get the name?

JR: We just had a bunch of names in mind and those were two that we just really loved. We wanted something whimsical, kind of adventurous, wanted to give her as many options as to how she wants to be known. And she just looked like a Wendylove when she came out.

TSF: Awesome. How have you dealt with releasing and promoting an album and touring all while trying to father a newborn? You must be exhausted.

JR: It?s funny, they draw on different energy, you know? You get ideas and you can?t spend six hours agonizing over them. I used to have time, so much time, you know? And now it?s not the same. You have blocks, periods of time, so at the end of the day you know very concretely what you?ve done. Which is great. Also it?s taught me how to decide to learn to calm down and appreciate things a little bit more than I have in the past. Like constant movement is not necessarily the end goal.

TSF: So you?ve still had to time to write since she was born?

JR: Yea definitely.

TSF: I?m sure your process has changed quite a bit.

JR: You know it hasn?t, since she?s been born. You know, the first three months of her life were very, they?re also very much the first three months of mine. And Haley?s. You know you?re learning how to do something you?ve never done before. But also you know we?re on the road together now and that?s work, that?s not writing work. You know, Haley?s writing, but it?s creative work that I think can only be fed by having a job. Really we?re on earth for one real reason and the rest is just kind of passing time, you know? And so with that reason there right in front of you, it does change you. I can?t say how exactly, it?s so early, but I do feel, I feel that it couldn?t help but change my writing in some ways. But I have no idea how yet.

TSF: What is it like having a newborn on tour with you?

JR: It?s awesome. It?s amazing. The bus has a?we fitted it so that there?s a bed in back and it?s got a crib. It seems to be pretty much like how it always has been except you know we?ve got a couple new people. It?s such a family already on the road, the people that I?m with. We?re really close and having them there, it?s great. So far it feels just terrific to come back after a show and I?ll go down and be a family. I believe this and I?ve believed this for a long time, I don?t believe because you choose to be a musician or choose to do something that you should have to give up having a stable family and a good relationship. And I think so far we?re really lucky that that appears to be true.

TSF: I want to talk a little bit about the new album. It?s been called a ?breakup album? but much of it seems influenced by your finding of new love, it?s quite hopeful. What do you think this record would be like if you didn?t find that new love when you did?

JR: Well I?d say that this was an interesting moment. I believe that with every record there?s a narrative that the record folds into. If a record is made how I believe they usually should be made, which is very regularly, you?re trying to encompass a certain amount of time, a certain amount of a number of preoccupations and get them all in that record so that when you move on, you have a record of that thing that happened. Whether it?s a writer?s block and adventure stories or kind of those ragged tumbling stuff. Those are all important moments for me, and they?re autobiographical in their own much less literal way. This was like a record where I began really in anger and real bitterness, and wanting to get some power back in my life. And I think I raised the pen at a moment when I think I needed to, where I felt like ?these aren?t the songs that I want. These are not going to be the songs that I?m going to look back on and feel proud of, and it?s not the story that I?m creating right now.?

It took a while for the real black rage that I had to kind of get hampered a little bit so the other good parts of my life could come out. And along that process I met Haley and things changed in like a really radical way for the better. And that wasn?t something that was planned, but it changed the way the record sounded and felt altogether.

TSF: Would you say this was your hardest record to write, the most challenging to write?

JR: Well, you know, I think it strangely was actually one of the easiest I?ve ever done. You know there was like, when I was realized that I wanted to write about this stuff in a way that had a little bit more?it was a bit more interesting to me, I started to realize you know there?s only a couple of moments really in your life when you can see yourself for what you are. I sort of imagine it?s dark down there and you can see which way the river is flowing only like in a flash of light. You know you see where the river is going, where it?s gonna end up. You know yourself really well for just a few seconds. And when you get that chance, to describe it is for me, that?s the part that?s easy. But to see it itself and see like ?New Lover? or ?Evil Eye? or ?Joy to You Baby,? you can actually see what it is you?re trying to describe. To try to describe a situation where you?re hopeful that someone?s doing better but you also hope that they?re doing worse. And that?s stuff that you have to sometimes get a flash of inspiration or flash of clarity. So really I think it was actually one of the easiest because it was very right in front of me and true in a really powerful way.

TSF: Was there any hesitation to release these more autobiographical songs? Was there ever a point where you tried to get back to writing less autobiographically like you had earlier, or was just it something you just felt you had to do?

JR: I just felt like this was?you know I was writing them, I just had to decide whether I wanted people to hear them. Not so much, not in regards to something like ?Joy to You Baby? or ?Lights,? which were more happy. But especially with the darker ones?but I feel like that?s the job, you know? If people don?t understand something, that?s fine. If they find it distasteful, that?s fine. But this is something that happened to me and I felt like if I didn?t write about it right now it?d be a betrayal of what I?m supposed to be doing. I mean there?s times where you?re just sticking around until that moment when something really important happens, or where you have something really important to say, and at that point, if you don?t say it then you?ve wasted all the time leading up to it. Plus I felt like if I did it right then it might be?people might be able to relate to it.

TSF: Before you started writing the album and before you went through the divorce, what direction were you expecting your next album after So Runs the World Away to take? Were you already expecting to strip it down a little bit or were you going to continue to do the more orchestral type of music?

JR: I thought about that a little bit. You know, when I?m finished with a record I usually don?t know where the next thing is gonna go, it takes a little while to know. And usually I?ve been pretty comfortable with that, but sometimes I thought in my mind like ?wow, what would have happened? What would have happened if I continued down the direction of So Runs?? I mean that was a direction that was like big and cool and comfortable, and I felt like I was saying big things, you know? But I don?t think you can continue in that direction. I also felt like maybe I?ve gone as far as I wanted to for now, in that style. And so, so, wind. I always think the wind was basically just knocked out of my sails and in this way it was really, really positive.

TSF: What is ?The Beast.? Where did the title come from?

JR: The Beast is the heartbreak. You can so easily make terrible decisions with your life. And I?m inclined to often make decisions that are rash, and that works out really good sometimes on stage, and other times in life it?s less good. But in a situation like coming back and it?s late-November, early-December [in] Brooklyn, and the sun comes up late and goes down early it?s?I was writing songs just to feel like I was not out there making bad mistakes and things that I would regret. So I thought of The Beast as being the heartbreak and if I could just kind of keep occupied with songs or whatever, it would help me to move forward in my life. And stay alive basically, until the page turned.

The IRS apologized Friday for what it acknowledged was "inappropriate" targeting of conservative political groups during the 2012 election to see whether they were violating their tax-exempt status. WASHINGTON (AP) ? The FBI is investigating potential civil rights violations at the Internal Revenue Service after the agency acknowledged the agency had singled out conservative groups for extra scrutiny, Attorney General Eric Holder said Wednesday.

Other potential crimes include making false statements to authorities and violating the Hatch Act, which prohibits federal employees from engaging in some partisan political activities, Holder said.

"I can assure you and the American people that we will take a dispassionate view of this," Holder told the House Judiciary Committee at a hearing Wednesday. "This will not be about parties, this will not be about ideological persuasions. Anybody who has broken the law will be held accountable."

But, Holder said, it will take time to determine if there was criminal wrongdoing.

Holder announced a day earlier that the Justice department had opened a criminal investigation, joining three committees in Congress that are looking into the matter. As the investigation widened, House Speaker John Boehner told reporters he had this question: "Who's going to jail over this scandal?"

"There are laws in place to prevent this type of abuse. Someone made a conscious decision to harass and to hold up these requests for tax exempt status," Boehner said. "I think we need to know who they are and whether they violated the law. Clearly someone violated the law."

Legal experts, however, said it could be difficult to prove that IRS officials or employees knowingly violated the civil rights of conservative groups. If there is a violation, the experts said, investigators can sometimes prove more easily that officials made false statements or obstructed justice in some other way.

"I think it's doubtful that any of these knuckleheads who engaged in the conduct that gave rise to this controversy knowingly believed that they were violating the law," said David H. Laufman, a former Justice Department lawyer. "But that remains to be seen. That's what investigations are for."

"It's more likely than not that," he said, "the conduct at issue here may constitute violations of IRS rules or standards or protocols or procedures but may fall short of what is necessary to constitute a criminal offense."

Even if IRS agents broke criminal laws in targeting conservative groups, investigators may have to prove they knowingly did it, a high standard, said Brian Galle, a former Justice Department lawyer who teaches law at Boston College.

"If the reason they were pursuing them was in order to punish them for their political activity, there might be a First Amendment concern there," Galle said. "On the other hand, if the reason that they were looking for tea party groups is because there had been press reports about this new group, the tea party, who was aimed primarily at getting more conservative people elected to office, then they were just responding to the evidence. It really depends on what their motives were."

Wednesday's hearing was the first of several in Congress that will focus on the issue.

The House Oversight Committee announced Wednesday that it would hold a hearing May 22, featuring Lois Lerner, the head of the IRS division that oversees tax exempt organizations, and former IRS Commissioner Douglas Shulman, whose five-year term ended in November.

The Senate Finance Committee announced a hearing for next Tuesday.

The House Ways and Means Committee will hold a hearing Friday, featuring the acting IRS commissioner, Steven Miller, and the Treasury inspector general for tax administration, J. Russell George.

At Wednesday's hearing, Rep. Jim Jordan, R-Ohio, said Lerner misled him and his staff when they asked her about complaints from conservative groups that they were being harassed by the IRS.

"I know for a fact, Lois Lerner lied to me, she lied to our personal staff, she lied to committee staff, she lied in correspondence," Jordan said.

Lerner learned about the targeting on June 29, 2011, according to a report Tuesday by the inspector general.

The report said ineffective management at the IRS allowed agents to improperly target tea party and other conservative groups for more than 18 months.

The report said that while their applications for tax exempt status languished, tea party groups were asked a host of inappropriate questions, including: Who are your donors? What are the political affiliations of officers? What issues are important to the organization, and what are your positions on those issues? Will any officers in the group run for public office? Where do you work?

The IRS started targeting groups with "Tea Party," ''Patriots" or "9/12 Project" in their applications for tax exempt status in March 2010, the inspector general's report said. By August 2010, it was part of the written criteria used to flag groups for additional scrutiny.

Colleen M. Kelley, president of the National Treasury Employees Union, said Wednesday that no union employees had been disciplined, as far as she knew. She noted that the IG's report said agents were not motivated by political bias.

Kelley told The Associated Press that low-level workers could not have specifically targeted conservative groups for long without the approval of supervisors. However, she noted, there are many levels of supervisors at the IRS.

"No processes or procedures or anything like that would ever be done just by front-line employees without any management involvement," Kelley said. "That's just not how it operates."

Also Wednesday, the IRS released a list of 179 advocacy groups that had been approved for tax-exempt status as of May 9. The list includes both seemingly conservative and progressive groups, including the Nevada County Tea Party Patriots and Progress Texas. A total of 34 included the words "tea party" in their names.

"Progress Texas and the tea party strongly disagree on the role of government," said a statement from Ed Espinoza, executive director of Progress Texas. "Yet, when we applied for tax-exempt status, Progress Texas received the same type of additional scrutiny that tea party groups are complaining about."

The IRS also pushed back against one aspect of the inspector general's report. The report said the chief counsel was briefed about conservative groups being targeted on Aug. 4, 2011.

The IRS, however, said in a statement that the meeting involved staff attorneys "several layers below" chief counsel William Wilkins.

"Wilkins did not learn about specific groups being singled out by name until earlier this year," the IRS statement said.

Galaxy S4 headed to NTT Docomo May 23, blue color confirmed

Blue Galaxy S4New color option for Japanese Galaxy S4

Japan's NTT Docomo will start selling the Samsung Galaxy S4 from next Thursday, May 23 -- and as per the pre-announcement rumors, it'll be getting a "blue arctic" color option in addition to "white frost" and "black mist." The Docomo version will also support digital terrestrial TV broadcasts through a pull-out antenna, and FeliCa NFC payment capabilities.

Besides that (and some branding changes) it's pretty much the same old Galaxy S4 we've come to know over the past few weeks. In case you were wondering, Japan's getting the quad-core Snapdragon 600 version most other countries are selling, as opposed to the rarer "octa-core" Exynos version.

YouTuber mappan777 has posted a quick side-by-side comparison between the new blue version and the better-known black model, which we've embedded past the break. (Quick spoiler: you're going to have a hard time telling these two colors apart.)

Newt Gingrich Can't Work Out What His Phone Is

"Here at Gingrich Productions, we've spent weeks figuring out," ponders Newt Gingrich. "What do you call this?" It's a phone, Newt. "But think about it. If it's taking pictures, it's not a cellphone. If you can get Wikipedia or go to Google, that's not a cellphone." Sigh.

New test for H7N9 bird flu in China may help slow outbreak, prevent pandemic

May 10, 2013 ? Breaking research appearing online today in Clinical Chemistry, the journal of AACC, demonstrates that a recently developed diagnostic test can detect the new strain of influenza (H7N9) currently causing an outbreak in China.

Since the end of March, 31 people have died from H7N9 infection, and the number of confirmed cases has climbed to 129. Evidence suggests that most H7N9 infections have arisen from poultry-to-human transmission, and that passage of the virus between humans is limited. However, researchers have also found mutations in the virus that are known to help avian viruses adapt to mammalian hosts. If these mutations lead to sustained human-to-human transmission, a serious pandemic could occur.

In this study, Wong et al. designed a diagnostic test with high specificity for the H7N9 virus that does not cross react with distantly related viruses, including all previously known avian and mammalian H7 viruses. They also show that this one-step quantitative real-time PCR assay enables specimen processing in about 3 hours.

According to the authors, this new test should also detect viruses closely related to the H7N9 virus. If confirmed, this capability could prove vital; it's likely that the H7N9 virus is evolving rapidly, and there could be multiple introductions of avian H7N9 viruses from animals to humans. The test also demonstrates a detection limit of ~0.04 median tissue culture infective dose (TCID50) per reaction. This means that it should be sensitive enough to identify patients with active virus replications.

"These results suggest that the established assay should be suitable for screening H7N9 viruses in human samples," said lead investigator Leo Poon, PhD, of the University of Hong Kong, though additional evaluation using clinical specimens from H7N9 patients is needed.

If validated, this diagnostic test could help health officials avert a potential pandemic by allowing them to monitor the spread of the virus. The test could also identify H7N9 patients in the early stages of infection, improving their chances of responding to clinical treatments.

IRS apologizes for singling out conservative groups: How did it happen?

The Internal Revenue Service singled out conservative political groups for additional scrutiny of their tax-exempt status during the 2012 election, which conservative groups say is proof that the IRS has been unfairly targeting them.

About 75 organizations with ?tea party? or ?patriot? in their tax-exemption applications received extra review during a broader screening process of political advocacy groups, said Lois Lerner, who oversees the tax-exemption division at the IRS.

?That was wrong. That was absolutely incorrect, it was insensitive, and it was inappropriate,? Ms. Lerner said Friday at a conference sponsored by the American Bar Association. ?That's not how we go about selecting cases for further review. The IRS would like to apologize for that.?

RECOMMENDED: What does the federal government do with your money? Take our taxes quiz.

Low-level employees in Cincinnati initiated the practice to deal with an influx of tax-exemption applications between 2010 and 2012, Lerner said.

?Mistakes were made initially, but they were in no way due to any political or partisan rationale,? said the IRS in a statement. ?We fixed the situation last year and have made significant progress in moving the centralized cases through our system.?

Conservative groups accused the IRS of targeting them during the 2012 election by sending additional questionnaires for their applications. So for them, the IRS apology is too little, too late.

?The IRS has demonstrated the most disturbing, illegal and outrageous abuse of government power,? Jenny Beth Martin, national coordinator for Tea Party Patriots, said in a statement. ?This deliberate targeting and harassment of Tea Party groups reaches a new low in illegal government activity and overreach.?

Top Republican lawmakers are calling for a White House investigation into the agency.

?Today, I call on the White House to conduct a transparent, government-wide review aimed at assuring the American people that these thuggish practices are not underway at the IRS or elsewhere in the administration against anyone, regardless of their political views,? said Sen. Mitch McConnell (R) of Kentucky in a statement.

Rep. Charles Boustany (R) of Louisiana told The Hill that he wanted the IRS to hand over the names of the employees involved with the matter.

?My greatest concern is what would have come from this blatant abuse of power if Ways and Means, as well as others, had not spoken up and stepped in to question the IRS about these activities,? said Representative Boustany, who brought up the issue in hearings of the Ways and Means Subcommittee on Oversight last year.

The IRS is an independent enforcement agency, with only two political appointees, said White House press secretary Jay Carney during a press conference Friday. The director during the time of the inappropriate activity ? former Commissioner Douglas Shulman ? was appointed by President George W. Bush.

?If this inappropriate activity did take place, the president would want it to be thoroughly investigated,? Mr. Carney said.

During the campaign, liberal groups thought that the IRS was not doing enough to review groups applying for tax-exempt status.

"That's the most interesting thing about this: They were actually doing it," Kenneth Gross, a campaign-finance law expert and former counsel of the Federal Election Commission, told ABC News.

The IRS apology could affect future regulations of political, tax-exempt groups, said Nick Nyhart, president and CEO of Public Campaign in Washington, which advocates campaign-finance reform.

"There are legitimate questions to be asked about political groups that are hiding behind a 501(c)4 status," Mr. Nyhart said in a statement. "It's unfortunate a few bad apples at the IRS will make it harder for those questions to be asked without claims of bias."

Dad's genome more ready at fertilization than mom's is -- but hers catches up

May 9, 2013 ? Researchers from Huntsman Cancer Institute (HCI) at the University of Utah have discovered that while the genes provided by the father arrive at fertilization pre-programmed to the state needed by the embryo, the genes provided by the mother are in a different state and must be reprogrammed to match. The findings have important implications for both developmental biology and cancer biology.

In the earliest stages, embryo cells have the potential to develop into any type of cell, a state called totipotency. Later, this potency becomes restricted through a process called differentiation. As a result, as cells continue to differentiate, they give rise to only a subset of the possible cell types.

"In cancer, normal processes of cell differentiation and growth go wrong, and cells either become arrested at an early state of differentiation, or instead go backwards and are 'reprogrammed' to become more like early embryo cells," said Bradley R. Cairns, co-author of the article and Senior Director of Basic Science at HCI. "By understanding how cells are normally programmed to the totipotent state, and how they develop from that totipotent state into specific cell types, we hope to better understand how cancer cells misregulate this process, and to use that knowledge to help us devise strategies to reverse this process." The research results will be published online as the cover story in the journal Cell on May 9.

Earlier work in the Cairns Lab showed that most genes important for guiding the early development of the embryo are already present in human sperm cells of the father in a "poised" state -- turned off, but with attached markers that make gene activation easy. "The logic is that all the important decision-making genes for early development are ready to go," said Cairns. "This poised state is never seen in fully differentiated cells such as skin cells."

In the current study, researchers in the Cairns Lab used high-throughput gene sequencing to comprehensively and precisely analyze DNA methylation patterns in the genomes of zebrafish, which is a common laboratory model both for developmental and cancer biology. Here, they examined egg cells, sperm cells, and four phases of embryonic development: three phases between fertilization and when the embryo's genome becomes active, and one phase after that point. Methylation -- in which molecules called methyl groups are selectively attached to certain areas of the DNA and turn off gene activity in those areas -- is one of the main markers of gene poising; poised genes lack DNA methylation, enabling gene activity later in embryo development.

Cairns' group found that the methylation pattern of the soon-to-differentiate embryo is identical to that of the sperm cell. In contrast, the pattern of the egg cell was initially quite different, but undergoes a striking set of changes to become exactly matched to that of the sperm DNA. Cairns' work suggests that egg DNA goes through this extensive reprogramming to prepare for the process of differentiation.

"The maternal genes that underwent DNA methylation reprogramming are among the most important loci for determining embryo development," said Cairns. "For example, many hox genes, which determine the body plan and also differentiation during hematopoiesis [the formation of blood cells], are methylated in the mother's genetic contribution and demethylated in the father's, and therefore, also in the embryo."

He said the work added another interesting finding. "We found that the mother's genome takes care of that remodeling on its own, without using the father's genome as a template." Cairns' experiments showed that when the father's genetic contribution was removed, the mother's genome still remodeled itself to the correct state.

"Basically, we're trying to understand how a single cell can make a decision to be any type of cell," said Cairns. "It is a fascinating fundamental question in biology that has implications for all aspects of development and many aspects of diseases such as cancer."

WebMD top management steps down

(Reuters) - WebMD Health Corp said Chief Executive Cavan Redmond, appointed last June, will be leaving as the health information provider reported a narrower quarterly loss.

Shares of the company were up about 10 percent at $27.94 in after-hours trading. They closed at $25.51 on the Nasdaq.

The company said its senior vice president of strategic and corporate development, David Schlanger, will serve as interim CEO.

WebMD, which operates medical websites providing health and disease related information, also said Anthony Vuolo will be replaced as chief financial officer by Peter Anevski, currently a senior vice-president of finance.

"The change announced today will best position us to build on the momentum that our senior management team has created to date," WebMD Chairman Martin Wygod said in a statement.

The popular website has struggled with a fall in advertising by budget-slashing pharmaceutical companies.

WebMD's first-quarter net loss narrowed by about 80 percent to $1.5 million, or 3 cents per share, from a loss of $7.8 million, or 14 cents per share, a year earlier.

Public portal advertising and sponsorship which makes up most of its revenue stream rose 6 percent to $93 million, while overall revenue rose 5 percent to $112.8 million.

(This version of the story corrects paragraph 8 to show that the total revenue figure was $112.8 million, not $112.7 million)
